I've heard "Your Majestry" in dramas and films a lot, so what difference does "HSM(His Serene Majesty)" have, especially "Serene"?

As an NES but not an English teacher:

I believe it is meant in the same way as: "with the greatest possible respect ....".
This normally precedes something such as: " I believe you are talking a lot of rubbish!" or similar.

"His Serene Majesty" was the style used by the Belgian royal family.
