Can you say:

Which city or state did you visit? Where and what did you like best of the US?

VS

Which city or state did you visit? Where and what did you like about the US?

Are both okay?

The first part is fine. I would use "in the US".
 
Like emsr2d2 I would use "in the US", but "about" is OK, too. I wouldn't use "of".
 
I'm happy with "about" when it's connected to "what did you like best ..." but the first word of that sentence is "Where", leading to "Where did you like best about the US?" and I find that very unnatural. If you want a word that fits "Where and what did you like best ...", I think only "in" fits.
