Is it a proper reply to the request? What do we usually say in response to the request like"Would/Do you mind doing sth?" if we agree to do it?

We don't have enough context to say whether that response is possible. If the person with the baby is somewhere else and is planning to bring himself/herself and the baby to the other person, the response could be correct with different punctuation.

Sarah: I have a favour to ask you.
Helen: What is it?
Sarah: I need some help. I'm thinking maybe I could drive us over to your place for the afternoon. Would you mind helping me take care of my baby?
Helen: Sure. Be careful on the way.

It's not a particularly natural dialogue but it does at least give a possible scenario in which it would work. However, bear in mind that "Sure" is the answer to the actual question and "Be careful on the way" is just some advice.

Maybe I didn't make my question clear. What I really want to know is whether 'sure' can be used to respond to the request in form of "would/do you mind...?" in some cases.

If you had written only that in post #1, it would have been a lot easier to answer.
