Is it correct to ask, "Who is the dialog between?"? For example:
A: Who is the dialog between?
B: It's between John and Jane.

Is it correct to ask no comma here "Who is the dialog between?"? For example:
A: Who is the dialog between?
B: It's between John and Jane.
It's OK, as long as you're talking about a conversation between two people in a film, a play or a TV show. I wouldn't use the word "dialogue" (that's the BrE spelling) in any other context.
