Using the verb to fulfill

If we want to express "The assumption is not valid for this problem", which sentence below can be used?

The assumption does not fulfill for this problem.
The assumption is not fulfilled for this problem.

You don't "fulfil for" something. It's fulfil + noun. (Note: fulfil is the BrE spelling, I believe fulfill (2 Ls at the end) is AmE.

As far as your actual question is concerned, I don't think that fulfil can be used for this example. You don't fulfil an assumption or a problem.

You fulfil an obligation.
You fulfil a promise.
You fulfil an order.

It means to bring something into effect, or to carry something out.

The assumption cannot be used for this problem.
The assumption is not relevant/suitable to this problem.

I tend to use AmE spelling and as a matter of fact, the spelling system of the usingenglish forum is happy with this too. Thank you for the additional information.

Of course you can. But they are two different things altogether.
You talk about fulfilling an assumption if it is valid to start with. Here it is not valid.
