Are the following both grammatical?

1. While I was reading the book, the story had me laughing out loud.
2. While I was reading the book, the story had me laugh out loud.

They're grammatical but neither is very natural. They make it sound as if the book and the story are two unconnected things. I see no reason for two clauses.

The story/book I was reading had me laughing out loud.
The story/book I was reading made me laugh out loud.

OK if in your examples you inserted, "........the story about _______"

Do you mean While I was reading the book, the story about had me laughing out loud?

that doesn't work for me at al.

Yankee probably meant, for example, something like "While I was reading the book, the story about a baby mimicking his father had me laughing/made me laugh ..."
