Peter has two brothers, but he doesn't speak to ………of them .
a-either
b-any
c-neither
d-both
what about c(neither)?

Not only that, but two is wrong.

May we also use "both of them" for the alternative?

No. In this negative construction, only either of them is possible.

Then, is it not possible to use the word "both" in negative construction?

Saying, "Peter has two brothers, and he doesn't speak to both of them" is ambiguous. If you say this, you will be asked to clarify.
