I don't want anyone to interrupt me no comma here when/while I am speaking.
They're both possible. I prefer "while" but bear in mind there's no need for anything after "interrupt me". The definition of "interrupt" is generally "to speak while someone else is speaking".

I don't want anyone to interrupt me.
