I would say "amount of sleep".

The length of time (hours of sleep) is singular and needs "the". And that is a run-on sentence.

Since "Hours of sleep" is one single factor, use the singular verb doesn't.

I disagree with Charlie. I would not use a singular verb after "hours of sleep".

However, I wouldn't use that phrase at all. What you mean is "quantity of sleep". Use that phrase.
 
I would use this structure.

It doesn't matter how many hours you sleep, it's the quality that counts.

It makes sense either way, but I would treat it as singular because you're making hours of sleep a thing.

You're essentially saying, "This doesn't matter, that does." Which one is more important: hours of sleep or sleep quality?
 
It doesn't matter how many hours you sleep. It is the quality that counts.

The quality of sleep takes precedence over hours slept.
 
It's not how long you sleep that matters. It's how well you sleep.
