In answer to "Could you please give me a ride?" I would say:

1) Sure, with pleasure.
2) Sure, it would be a pleasure.

I definitely wouldn't say "Sure, it's a pleasure". However, I might say "Sure, it's always a pleasure to have some company on a car journey."
