I assume that the reference to "Columbus" means to discover/find/explore. However, it is a very strange sentence.

What is the source of this sentence, LeTyan?

Its source is unknown. It was used as an example for learning iversion grammar in a textbook. However there is no citation of that.

Every one of us has in him a continent of undiscovered character. Blessed is he who acts the Columbus to his own soul.

It's apparently from a book called 'Words of Life' by Charles L Wallis

So that doesn't seem like a regular way of speaking English. It's more like a literature thing right? Maybe it is Okay that non-professionals don't understand what it means?

No one said that they didn't understand it. Mike explained the meaning. Columbus was an explorer who found a new world. A person who seeks and finds new parts of his/he character is like Columbus in that he/she is also an explorer.
