My suggested edits:

Dear Sir,
I would like to call you in order to ask a few questions regarding the e-mail that I received yesterday from the Human Resources Specialist.

Please let me know when it would be convenient for you.

Thanks in advance,
(Your Name)

very nice...

My suggestion:

Dear [RECEIVER'S NAME],

I would like to discuss with you regarding the E-mail which was sent to me by the 'Human Resources Specialist' yesterday; my preference goes to have a phone conversation.

I will appreciate it indeed if you let me know that according to your convenience, when we can discuss about it.

Best regards / [YOURNAME].

Hello Mega,
I'm sorry, but this is not natural. The suggestions give prior to yours is fine. The parts I've put into italics are problems in your suggestion.
