What context do you mean? You said the contex. Do have any particular context in mind?

Generally, if John has two brothers, and one of them is older and the other one is younger than John, then the word "brother" alone doesn't tell us which brother is talked about.

I think we can't know that unless the speaker wants to add the 'big/older' or 'little/younger' to this word.
 
John will go to Singapore to visit his friends, while his brother, Jim, will be home to look after their parents.
