Should I use "a best friend" or "the best friend" should it be "her" instead of "she" and is "in others" correct?

Context: I am talking about my friend.

"I had a best friend at the university. She and I were very different in some ways, but very similar in others."
 
Use “she” and “in others” in your second sentence.
