Verb to match the subject "number,' not the object of the preposition. Third grade stuff.

Take out the "of animals." Prepositional phrases don't count. Therefore, the answer is "is."

Swan: 'A large number of' is used before plurals and the a following verb is PLURAL. Therefore, 'are' should be used.
